High end microsystems are widely used in innovative research in related frontier fields such as biology and basic medicine, especially the 10-100nm super-resolution micro optical imaging technology, which plays an irreplaceable role in current biology and basic medicine research.
As a necessary tool for biomedical experimental research, laser scanning confocal microscope has higher resolution than traditional fluorescence microscope, and can perform tomographic scanning 3D imaging. However, the thickness of the sample that can be observed by the confocal microscope is generally less than 100 um, and a two-photon microscope is required to observe a deeper sample. The great advantage of two-photon microscopy * is the depth of observation.
However, neither laser scanning confocal microscope nor two-photon microscope can get rid of the diffraction limit. In order to further explore the microscopic world, a microscope with higher resolution is needed. The STED microscope came into being. It introduced the loss beam to further compress the fluorescent spot on the basis of the confocal microscope, so as to realize super-resolution imaging.
